Studies on Relationship between Polynuclear Aromatic Hydrocarbon Concentration and Genotoxic Effects of Total Suspended Particulate Extract



Abstract
The total suspended particulate(T.S.P.) were collected on quartz fiber from Mar., 1988 to Feb., 1989 at Chung Ang University using Andersen high-volume air sampler, polynuclear aromatic hydrocarbon(PAH) concentrations in these particulates were analyzed by high performance liquid chromatograph. And the clastogenic effects of airborne particle extracts were examined by the bone marrow micronucleus test.
1) The annual arithmetic mean concentration of T.S.P.(1988.3¡­1989.2) was 93.19§¶ / §©.
2) The concentration of T.S.P. was low in fall and summer, high in winter and spring.
3) The annual arithmetic mean concentration of polynuclear aromatic hydrocarbon(PAH) were ; benzo(a)pyrene 1.14 ng / §©, benzo(ghi)perylene 1.27 ng / §©, benzo(k)fluoranthene 0.54 ng / §©.
And the cytogenic effects, in vivo, of the standard benzo(a)pyrene and were studied adult male mice by the micronucleus test.
4) The lowest effective dose was 25 mg / kg with standard benzo(a)pyrene and had a dose-response between 25 and 200 mg / kg body weight.
5) In the micronucleus test, the mutagenicity by the organic matter extracted(tar) had a clastogenic effect with dose-response between 30, 60, 120 and 300 mg / kg body weight by the intraperitoneal injection and, in the seasonal variation, frequency of micronucleated polychromatic erythrocyte(MNPCE) was high in winter, low in summer.
6) The relationship between frequency of MNPCE and P.A.H. concentration (Benzo(a)pyrene, Benzo(ghi)perylene, Benzo(k)fluorantehne.) in T.S.P. was high.
